I want to Send SMS using flowroute API. I want to use PHP or Laravel Framework. I didn't get any good tutorial about it.
You know, that you just need to read and click a littlebit? The address you refered to, shows PHP-examples. On the target-github-page you can find the text "To work with the SDK, please read and follow the steps in the Flowroute PHP Library v2.". What is the problem?
I am stuck in a situation in which when a user registers, he/she get a confirmation SMS on his/her mobile number, and he/she able to confirm their application by replying to the confirmation SMS, be repling like YES/NO to that confirmation sms..
Is there any SMS gateway available which provides the suitable functionality, I mean an API which takes user input from sms reply..

Twilio has the ability to receive sms messages.

There is a definite solution available for this. Try exploring with FrontLine SMS that offers a similar functionality, though you will have to then use a 10 digit registered number to send your confirmation SMS.
How to use frontline sms:
Get an internet dongle from a cellular company (Idea, Airtel,etc)
connect dongle to PC
install frontline SMS
Frontline SMS detects dongle.
Frontline SMS API's available to read incoming SMS on dongle and forward to your server with POST/GET methods.

I have done it using tables.
We can not use internal or external CSS in email. We can use the inline CSS. But in html email it's best to use tables.
I wanted to create (or at least learn/know how it is done) application(or configuration?) that does similar to what craigslist does when people choose to hide their email with the "anonymous" option when making posts. I suspect that it is done with what's called email relaying. I'd like to find out how it is done in process - from when user enter their email to receiving an email via an anonymous email address. I come from a "LAMP" background at an intermediate level so please bear with me and kindly explain.

The easiest way to do this is to receive email with your php app. There are a wide range of ways to do this, collecting using cron, piping from an email server directly into your app or using a third party like CloudMailin.
