Trying to work out a set of .htaccess mod_rewrite rules that will set these links:
domain.com/content-page
domain.com/region/another-page
domain.com/region/brand/more-content-here
to fetch files:
domain.com/content-page.php
domain.com/region-another-page.php
domain.com/region-brand-more-content-here.php
Where 'region' and 'brand' and the page name are all variable/changeable.
So far I have:

This works but only for one forward slash. As mentioned above, URLs after the domain may have none, one or two forward slashes. Haven't had any luck trying to extend this to deal with multiple (or not) slashes.

It depends on where you put the rule for your rewrite. The examples you provided do work. But you have to put them before your last htaccess line.
RewriteCond $1 !^(index\.php|resources|robots\.txt)

This will create the proper output, and can be tested through something like a htaccess tester
If you want it specifically for just that url you could resort to:
RewriteCond $1 !^(index\.php|resources|robots\.txt)
RewriteCond %{REQUEST_FILENAME} !-f
RewriteCond %{REQUEST_FILENAME} !-d
RewriteRule ^mysite/loginPage/$ mysite/loginPage? [R=301,L]

You could also specify an absolute redirect with
RewriteRule ^mysite/loginPage/$ http://%{HTTP_HOST}/mysite/loginPage? [R=301,L]

In that case, your rule should just match specifically this URI rather than .*/.* etc.

You don't really need QSA flag in last rule also.
Make sure to clear your browser cache before testing this change.
I need to 301 redirect all URLs ending in .php to the non extension version. But the rewrite rules need to work with the existing ones below.

Tried a few various ideas but they didn't seem to work in with the existing rules and htaccess is not my strength. Also need it to be querystring friendly. Any help would be appreciated.

The first page works as /polls/username but the second doesn't read the poll value and I think redirects to polls.php.
Could anyone explain what I'm doing wrong?

Here, I've swapped your two rules around, so we check for IDs first (for getpoll - note how I have removed letters from the expression). If we're not doing an ID check (ie, there are letters in the segment), then it'll rewrite for polls.

Per arco444, if the username is a number, this wouldn't work. This is a more specific solution:

EDIT: commented the R=301 parts because, as explained in a comment:
Be careful with that R=301! Having it there makes many browsers cache the .htaccess-file indefinitely: It somehow becomes irreversible if you can't clear the browser-cache on all machines that opened it. When testing, better go with simple R or R=302
After you've completed your tests, you can use R=301.
To complement Jon Lin's answer, here is a no-trailing-slash technique that also works if the website is located in a directory (like example.org/blog/):
RewriteCond %{REQUEST_FILENAME} !-d
RewriteCond %{REQUEST_URI} (.+)/$
RewriteRule ^ %1 [R=301,L]
For the sake of completeness, here is an alternative emphasizing that REQUEST_URI starts with a slash (at least in .htaccess files):

Just don't use %{REQUEST_URI} (.*)/$. Because in the root directory REQUEST_URI equals /, the leading slash, and it would be misinterpreted as a trailing slash.

This removes html code or php if you supplement it. Allows you to add trailing slash and it come up as well as the url without the trailing slash all bypassing the 404 code. Plus a little added security.
I want to redirect what's in link after localhost/myScript/ to localhost/myScript/test.php?var=$1.
So.. something like

(with or without the last slash) would redirect to
localhost/myScript/test.php?var=this/is/just/a/test
This is what I've got, but it's not good enough.

But, I get $_GET['var'] = 'test.php'.
I see 2 main approaches:
1. Two separate rules to deal with URL with and without trailing slash. It will only rewrite requests to non-existing files/folder thus preventing rewrite loop you are having:
