I want to add public/index.php to all request using .htaccess
I tried like this
RewriteEngine on
RewriteRule ^/(.*)$ /public/index.php/$1 [NC,L]
but it didn't changed ..
Example :
If i type
localhost/project/first
it should request
localhost/project/public/index.php/first
What is the issue in my .htaccess ?
You can use a rule like this in /project/.htaccess:

RewriteCond are required to avoid rewriting for real files and directories. And There shouldn't be any leading slash in RewriteRule pattern in .htaccess. .htaccess is per directory directive and Apache strips the current directory path (thus leading slash) from RewriteRule URI pattern.

I have a route that expects an id:

But I need the URL the user sees to be friendly as:

I thought something like this would work, but I get a 404:

The user sees the pretty url, but you are not interested in what the seo-title is. What you are interested in is the number that is hidden in it. As the number is in the very back of the pretty url, let's just match on that:
RewriteRule ^[^/]+-([0-9]+)\.html detalhe/$1 [L]
If you are trying to use another rewrite that will not be routed through the default MVC route index.php then you need to make sure you place your new rewrite before those rules. Also that folder you are redirecting to will need to have a index PHP file to do something as well.

I am using .htaccess to manage clean URL's but for some reason I get a 404 Not Found Apache error in some cases.
If a user goes to http://domain.com/profile/ everything is fine.
If a user goes to http://domain.com/profile they will get the error

You can redirect the url with the url which has trailing slash using 301 redirect
Try the following along with your rewrite condition. This will redirect any url request to url with trailing slash. ex. http://domain.com/profile will be redirected to http://domain.com/profile/

I would also like to force the site to have the 'www' subdomain and most importunately add a trailing slash if the url doesn't have one.
I am an absolute noob with mod_rewrite and I've tried accomplishing this on my own by combining other code I found on google (sad I know), but I always end up with a 500 error.

Try separating out the www and the trailing slash check. This is tested and hopefully working for you. You didn't say if you're running placing at domain root or in a subdirectory - usually good info when asking for help with htaccess.
