I just don't understand GMT on how it works with your php sql server time. How do you relate to your database with the minutes offset? So if this produces -420 minutes how do we relate that to our database time (for example unix time 2016-09-14 22:40:31)(your NOW() time)?
I am just not sure how to implement this in PHP? There is some vague information I have found on how to use it after you get your minutes offset.

You can use the bellow code to set the time zone:
date_default_timezone_set('Europe/London');
This code will add +1 hour onto the timestamp.
$timestamp= strtotime("+1 hour");

You can convert a specific offset to a DateTimeZone:
$offset = '-0500';
$isDST = 1; // Daylight Saving 1 - on, 0 - off
$timezoneName = timezone_name_from_abbr('', intval($offset, 10) * 36, $isDST);
$timezone = new DateTimeZone($timezoneName);
Then you can use it in a DateTime constructor, e.g.
$datetime = new DateTime('2012-04-21 01:13:30', $timezone);
or with the setter:
$datetime->setTimezone($timezone);
In the latter case, if $datetime was constructed with a different timezone, the date/time will be converted to specified timezone.
How can I compute time difference in PHP?
example: 2:00 and 3:30.
I want to convert the time to seconds then subtract them then convert it back to hours and minutes to know the difference. Is there an easier way to get the difference?
Look at the PHP DateTime object.
$dateA = new DateTime('2:00');
$dateB = new DateTime('3:00');
$difference = $dateA->diff($dateB);
(assuming you have >= PHP 5.3)
You can also do it the procedural way...

You can get the hour offset like so...
$hours = $difference / 3600;
If you are dealing with times that fall between a 24 hour period (0:00 - 23:59), you could also do...
$hours = (int) date('g', $difference);
Though that is probably too inflexible to be worth implementing.

I used this to calculate the difference between server time and the users local time. Grab the hour difference and drop that in a form when the user is registering. I then use it to update the time on the site for the user when they do stuff online.
Once I got it working, I switched this line ...

Now I can compare the user time and the server time! You can grab the seconds and mins as well.
I have a table which shows the time since a job was raised.

$difference needs to exclude any time outside of business hours (ex, 9-5 and weekends).
How could I tackle this?
The thing you have to do are 3 in numbers.
You take your start date and calculate the rest time on this day (if it is a business day)
You take your end date and calulate the time on this day and
you take the days in between and multiply them with your business hours (just those, that are business days)
And with that you are done.
Find a little class attached, which does those things. Be aware that there is no error handling, time zone settings, daylight saving time, ...

Use DateTime.
Using UNIX time for this is slightly absurd, and you would have to literally remake DateTime.
Look up relative formats where you can specify the hour on the day, e.g.
$date = new DateTime($raised);
$business_start = new DateTime("09:00"); // 9am today
$business_end = new DateTime("17:00"); // 5pm today
The rest is for you to work out.
Oh, and instead of start/end, you could probably use DateInterval with a value of P8H ("period 8 hours")
The problem with using timestamps directly is that you are assigning a context to a counter of seconds. You have to work backwards from the times you want to exclude and work out their timestamps beforehand. You might want to try redesigning your storage of when a job is raised. Maybe set an expiry time for it instead?

You may want to take a look at the phpjs site. They have code showing how a substantial number of PHP functions can be done in JS.
Specifically: strtotime and date
JS doesn't have anything remotely close to strtotime. You'd have to determine "next tuesday" yourself. Once you've got that, you can extract a timestamp value using .getTime(), which will be the number of milliseconds since Jan 1/1970. This value can also be fed back into a new date object as a parameter, so you can do date math using simple numbers externally, then create a new date object again using the result.
e.g.
var now = new Date();
var ts = now.getTime();
var next_week = ts + (86400 * 7 * 1000);
next_week_object = new Date(next_week);
Once you've got the "next tuesday" code figured out, the rest is trivial
To get milliseconds till the next tuesday (nearest in the future):

In PHP, you can tell if a given date is during the Daylight Savings Time period by using something like this:
$isDST = date("I", $myDate); // 1 or 0
The problem is that this only tells you whether that one point in time is in DST. Is there a reliable way to check whether DST is in effect at any time in that timezone?
Edit to clarify:
Brisbane, Australia does not observe daylight savings at any time of the year. All year around, it is GMT+10.
Sydney, Australia does, from October to March when it changes from GMT+10 to GMT+11.
I'm wondering if there would be some existing method, or a way to implement a method which works as such:

The getTransitions() function gives you information about each time the offset changes for a timezone. This includes historical data (Brisbane had daylight savings in 1916.. who knew?), so this function checks if there's an offset change in the future or not.

DateTimeZone::getTransitions might help.
You could probably wing it:
$hasDst = date("I", strtotime('June 1')) !== date("I", strtotime('Jan 1'));
Otherwise you'd need to parse the text-based zoneinfo data files.
I don't think so, but since almost every country that observes DST changes its time for an entire season or two, you could try to test 4 points during any given year.
For example, test date("I", $date) for 2009/01/01, 2009/04/01, 2009/07/01 and 2009/10/01. If that timezone falls into DST, then at least one of those dates will return 1.
date has to be on the user/server timezone for it to work, and you can't use a range with date as you do with getTransitions
